Three Matching Rules

  1. The packing must match the analytical column— same bonded phase, same particle size, ideally the same batch. Using a different packing in the guard column is equivalent to putting a short column of different selectivity in front of the analytical column, which changes retention and selectivity.
  2. Match the internal diameter to the analytical column— a mismatch creates an abrupt change in the flow path at the connection, which directly causes band broadening and tailing.
  3. Keep the length as short as possible— typically 5–20 mm. The guard column itself contributes retention and extra-column volume: the longer it is, the better the protection and the greater the loss in efficiency.
Guard column vs. in-line filter: an in-line filter (0.2–2 µm frit) intercepts particulates only and adds almost no dead volume; a guard column intercepts both particulatesand strongly adsorbed components, but adds dead volume. If the sample is clean with only occasional particulates, use an in-line filter; if the matrix is complex (plasma, fermentation broth, herbal extracts), use a guard column.

Two details when replacing the cartridge: (1) install the guard column in the same flow direction as the analytical column — reversing it flushes contaminants trapped at the inlet back into the analytical column; (2) re-equilibrate and re-run system suitability afterwards — a cartridge change is a modification that affects retention, and in a GMP setting the lot number and replacement date should be recorded.
